Characterization of Argonaute nucleases from mesophilic bacteria Paenibacillus borealis and Brevibacillus laterosporus

Abstract Thermophilic Argonaute proteins (Agos) have been shown to utilize small DNA guides for cleaving complementary in vitro, which shows great potential nucleic acid detection. In this study, we explored mesophilic Agos the detection of molecule by cooperating with allosteric transcription factors (aTFs). Two from bacteria, Paenibacillus borealis ( Pb Ago) and Brevibacillus laterosporus Bl Ago), showed nuclease activity single-stranded at moderate temperatures (37 °C) using 5′-phosphorylated 5′-hydroxylated guides. Both perform programmable cleavage double-stranded DNA, especially AT-rich regions plasmid. Furthermore, developed a simple low-cost p-hydroxybenzoic method based on DNA-guided effect HosA, expands application Agos.
